Discovery and biological effects of cisplatin and metallocene compounds and their application in anticancer chemotherapy

Abstract: This paper reviews the mode of action of cisplatin on tumor cells, the brief prospect of metallocene compounds as antitumor drugs and the application prospect of metallocene compounds in antitumor chemotherapy. The molecular mechanism of the interaction between cisplatin and DNA, DNA repair mechanism and cellular proteins were discussed. The molecular background of cisplatin sensitivity and drug resistance and its effect on antitumor immune response were evaluated. In addition, some metallocene metals with high antitumor activity (titanocene, vanadium, molybdenum, ferrocene and zirconium) are also reviewed in this paper.
