Synthesis, characterization and antibacterial study of PD (II) and Pt (II) complexes of some diaminopyrimidine derivatives

Abstract: Palladium (ⅱ) and platinum (ⅱ) complexes of trimethoprim and pyrimidine were synthesized and characterized by elemental analysis, UV-Vis, IR and NMR. These complexes are tetra coordinated square planar species, containing two drug molecules and two chloride ions or thiocyanate ions. Spectral analysis confirmed the coordination between metal ions and pyrimidine nitrogen atoms. The antibacterial activity of the complex was screened. They showed different activities, and the active metal complexes showed stronger inhibition than trimethoprim or pyrimidine. Pyrimidine palladium (ⅱ) complex has unique inhibitory activity against Pseudomonas aeruginosa and Bifidobacterium brevis, and other complexes have no inhibitory activity or the drugs showed any activity against these bacteria isolates. The MIC and MBC determinations revealed that these Pd(II) complexes are the most active. Structure activity relationship showed that Pt(II) complexes containing chloride ions are more active, while for Pd(II) complexes containing thiocyanate ions showed more enhanced activity than those containing chloride ions.
