Abstract: The continuous change of protein conformation caused by interaction with isoflurane may be a new molecular aspect of pretreatment. We pre incubated human serum albumin with isoflurane, dialyzed to the release agent, and evaluated the protein conformation. The sensitivity of methylglyoxal and nitrophenylacetic acid to chemical modification was also studied. Isoflurane has a lasting effect on protein conformation. The increased sensitivity of surface residues to chemical modification is related to this change in conformation. The modification of isoflurane treated HSA includes cross-linking within and between subunits, which may be the result of anesthesia induced changes in polymer subsets. This irreversible effect of isoflurane may be a mechanism of pretreatment.
