Abstract: Matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) remodel the cell surrounding environment by regulating extracellular matrix proteins, cell surface components, neurotransmitter receptors and the cleavage of growth factors that regulate cell adhesion, synaptic formation, synaptic plasticity and long-term enhancement. Interestingly, the increase of MMP activity and the imbalance between MMP and TIMP are also related to various pathological conditions. In this paper, we discuss various animal models, which show that the activation of gelatinases MMP-2 and MMP-9 is involved in the pathogenesis of drug-dependent Alzheimer s disease; Disease and epilepsy.
